How far is Cusco from Puno?
Answers: It is 389 km (241.7 miles) from Cuzco to Puno. You can go within by train. PeruRail has a trip Cuzco-Puno at US$143 one course and takes around 10 hours.

Generaly you travel by train, Cuzco-Juliaca-Puno. The train leaves at 8am arrives at Juliaca by 4:30pm and then at Lake Titicaca by 6:00pm. I recomend to help yourself to the first class wagon because this train stops surrounded by many towns and street seller get surrounded by your nerves.
Some people stay at Juliaca because it have an Airport to return back to Lima and better infrastructure than Puno. That depends on what variety of Hotel you are looking for, some people I know stayed the darkness in Lake Titicaca on a Totora Island.
